Sheep blood agar is a highly nutritive medium and supplementation of the sheep blood (5 -10%) provides additional growth factors and also serves on the basis of determining haemolytic reaction.
It is a selective and differential medium for cultivation of enteric Microorganisms from a variety of clinical specimens. Gram negative bacteria usually grow well on the medium and are differentiated their ability to ferment lactose.
The selective action of this medium is attributed to bile salts which is inhibitory to most species of gram positive bacteria.
Kirby Bauer et al., recommended this medium for performing antibiotic susceptibility test using a single disc of high concentration of antimicrobial agent and the zone diameters observed.

The cultivation medium for gonococci should ideally be a rich nutrients base with
blood, either partially lysed or completely lysed.

This medium was originally
designed for the isolation of
V.cholerae and V. parahaemolyticus, most Vibrios grow to healthy large colonies with many different colonial morphologies. TCBS Agar is also recommended by APHA for the selective isolation of V.cholerae and V.parahaemolyticus.
Mueller Hinton Agar supplemented with 5% defibrinated sheep blood, and details both the quality control procedures and interpretive criteria for use with S. pneumoniae and other Streptococci.
Methylene blue and Eosin-Y
inhibit gram-positive bacteria and serve as differential
indicators in response to the fermentation of carbohydrates. The coliforms produce purplish black colonies due to taking up of methylene blue-eosin dye complex, when the pH drops. The dye complex is absorbed into the colony.
Mannitol Salt Agar is used as
a selective media for the isolation of pathogenic Staphylococci from clinical and non-clinical samples.
Cooked Meat Medium (R.C. Medium) is used for cultivation of aerobes and anaerobes, especially pathogenic Clostridia from
clinical, food and water samples. This can also be used as a maintenance medium for stock cultures.
Single strength MacConkey Broth (Single and Double strength) w/Neutral Red is used for primary isolation of coliforms from large samples such as water or waste water.
Double strength MacConkey Broth (Single and Double strength) w/Neutral Red is used for primary isolation of coliforms from large samples such as water or waste water.
Tryptone Broth is used for the detection of indole production by coliforms.
Mannitol Motility Test Medium is a semisolid medium suitable for determining motility and mannitol fermentation.
Urea Agar Base with the addition of Urea is recommended for the detection of urease production, particularly by members of the genus Proteus.
